Introduction:Tamoxifen, also known as Nolvadex, is a widely used breast cancer drug that works by blocking estrogen receptors in breast tissue. The drug is used in postmenopausal women who have been or may be at an increased risk of developing breast cancer. Tamoxifen is available as a pill and oral solution.Objectives:To evaluate the efficacy and safety of tamoxifen in postmenopausal women with hormone receptor-positive advanced breast cancer.Methods:This is a prospective, multicenter study that enrolled 133 postmenopausal women with early breast cancer who had been treated with tamoxifen for a minimum of one year. The study enrolled a total of 136 women. The patients had a median age of 75 years (range, 25 to 100 years). Patients with advanced breast cancer were treated for 10 years with tamoxifen. The patients were divided into 3 groups based on the diagnosis of breast cancer: Group 1, patients treated with tamoxifen for 10 years had a lower risk of developing breast cancer; Group 2, patients treated with tamoxifen for 5 years were more likely to develop breast cancer than Group 3 patients.Results:In both groups, the overall 5-year survival rate was similar in both groups. The median follow-up was 34 months (range, 13 to 72 months). Although Group 1 patients had a lower 5-year survival rate than Group 2 patients, the hazard ratio for overall survival was significantly higher in Group 1 patients treated with tamoxifen than in patients treated with tamoxifen for 5 years (HR, 1.17, 95% CI, 1.02 to 1.39; p=0.04). The hazard ratio for tamoxifen-associated complications was 0.80 (95% CI, 0.55 to 1.29; p=0.19).Conclusions:Tamoxifen is a safe and well-tolerated treatment for postmenopausal women with advanced breast cancer. Tamoxifen may be a potential alternative to the standard tamoxifen regimen in postmenopausal women with breast cancer. Although tamoxifen has fewer adverse drug reactions, it is generally well-tolerated.
